Astragaloside IV and calycosin 7-O-β-D-glucoside are useful quality markers for Astragali Radix and can support chromatographic characterization of the extract profile. For procurement comparison, extract ratio, marker percentage, and analytical method should remain separate fields rather than being treated as equivalent expressions of extract strength.

Technical & Quality Considerations

Analytical evaluation should establish botanical identity first and then verify the specification system being purchased. Extract ratio is not a purity measurement, and TLC identity testing should not be presented as proof of concentration. Standardized grades require methods appropriate to the defined target, while chromatographic profiling can provide additional information about the relationship between saponin and isoflavonoid fractions.

Physical behavior changes with extract composition. Polysaccharide-rich materials are generally more relevant to aqueous dispersion systems, while increasing astragaloside enrichment changes the balance of the botanical matrix. Astragaloside IV itself has poor aqueous solubility, making dissolution and dispersion especially important in liquid applications. Color, sensory profile, wetting, and sedimentation should therefore be evaluated by extract type.

For dry-product scale-up, R&D should assess powder flow, premixing strategy, blend uniformity, capsule filling, and tablet compression. Beverage and reconstituted powder systems require additional evaluation of wetting rate, dispersion, haze, sediment formation, flavor impact, and process compatibility. Ratio, polysaccharide-standardized, and astragaloside-standardized extracts should be treated as different formulation matrices rather than interchangeable powder grades.
